Course aims
To provide knowledge about the most important steps of molecular cloning and expression in prokaryotes.
Brief description of the course
Cloning steps and components, restriction enzymes, cDNA synthesis, gene synthesis, polymerase chain reaction (PCR), primer design, RNA and DNA analysis, DNA sequencing, BLAST, transformation, fusion protein expression, purification and analysis, peptide analysis and concentration determination.
Learning outcomes in the course
Upon completing the course the student:
- is able to design a primer and can practically carry out the main steps of molecular cloning;
- is able to find and analyze potential sources of error in the cloning process;
- can express and purify fusion proteins on a large scale using affinity chromatography and analyze the process.
